意味としては欠けているものを嘆くのでは無く、ポジティブに考えよう、みたいな意味合いだったと思うのです。アメリカのドーナツ屋の広告だったような気もしますが、英文・日本語などでいろいろ検索しても、これといった回答がでてきません。

upon と on の違いはありますけど、こんな感じっぽいです。

Twixt は、betwixt の縮約系で、betwixt は、古語で between の意味だそうです。
